No, you need the tube, or the lipstick bullet will be unprotected. The refill is only $6 less than the full lipstick, so you may as well buy the full lipstick and then you can get the refill next time and use the tube :)

You cannot just use the refill as a lipstick, as there really isn't any protection around the lipstick to protect it from getting beaten up, melted, smooshed and broken over time. Unless you never carry your lip products around, you should buy the full product. Also, you can't roll the product up and down, and so the product is soft enough that it will break off and you'll lose the majority of the product after one use.
Ultimately, not practical to buy just the refill.

I suppose that you could, but you would have to find something to hold it with. The case is the holder so unless you put it in some sort of container, I don't know how you would hold it to apply it. The refill usually snaps into place inside the old lipstick case; you hear a click and it is then installed.
